Proteins difference of Endler's guppy / Poecilia reticulata.

The general aim of our research work is undestending of proteins difference between guppy and Endler's.
We have a doubt that these are different species of fishes becouse we have all resultes of crosses indicate accessories these fish to one mind - we have seen splitting for reference forms of F2 that no one interspecific hybrids do not occur.

Also we have results of research work other lab in USA:

Their resultes - no difference also.

Our research work was performed in Insitute of General Genetics by N. I. Vavilov RAS.

Groups of fishes:

1. MOSСOW RED (Russian breed) - 4 pcs.
2. GULFSTREAM (Russian breed)- 4 pcs.
3. WILD FERAL GUPPY FROM MOSCOW-RIVER - 4 pcs.
4. Poecilia wingei х Poecilia reticulata hybrid - 8 pcs.
5. ENDLER'S guppy - 7 pcs.
6. MOLLY (control) - 1pcs.

Were studied 13 enzymes isolated from the liver and muscles - and 20 loci (alleles) - one enzyme heve 1 to 3 or more allelic variants (isoforms).

So, the result of protein electrophoresis:

we found 3 enzimes and, respectively, and 4 alleles (isoforms) with differences between Endler's guppy and all groups of Poecilia reticulata (wild and breeds), which allows previously attributed these different species of fish.

We also agreed in September to see more fishes (10-15 fishes in each group) for three selected enzymes. I hope that I will receive wild guppy from Colombia in September, by which time I will catch wild guppy in Moscow River, Moscow's Red guppy and Endler's guppy.

In addition, the fall we begin to study the DNA (as well as chromosomes) of these fish. Comparing all the results will be made definitive conclusions...

We analyzed several species of fish again (36 fishes) - Poecilia sp. (P.reiculata, P.wingei, wingei x reticulata hybrids, P.velifera) and Xiphophorus sp. (X.helleri, X. maculatus) as well as among themselves.

We found 3 isoforms of enzimes and, respectively with differences between Endler's guppy and all groups of Poecilia reticulata (2 wild populations, 2 breeds and few hybrids).

The difference from the Endler's guppy (P. wingei):

Endler's guppy / Colombia wild - the difference of 3 isoforms of enzymes (hereinafter I will write count of differences only)

Endler's guppy / Moscow-River population - 3

Moscow-river population / Columbia wild - 2

X. helleri green / X.maculatus (two breeds) - 7-8

X. maculatus yellow/ X.maculatus black - 3

P. velifera / X. helleri green - 6

P. veifera / X. maculatus (two breeds)- 7-8

P.velifera / Endler's guppy (or Moscow-river population or Columbia wild) - 7

X. helleri green / Moscow-river population (or Columbia wild) - 8

Moscow guppy / Non-Moscow guppy (Italian breeding) - 2

Poecilia sp. differed from each other by 1-7 enzymes.

But, between Endler's guppy, all breeds and populations of guppies difference was 1-3, and only with P. velifera it was 7.

Two wild populations of guppies (Columbia and Moscow populations) showed a difference between to 2 enzymes, and between Endler's guppy and each of these populations - the difference was 3 enzyme. In this case, the difference between Moscow guppies and Italian non-Moscow breed was 2 enzymes.

Xiphophorus sp. differed from each other by 4-8 enzymes.

At the same time for comparison Molly differed from all the guppies and Endler and 7 enzymes.

Thus, representatives of the Poecilia sp. and Xiphophorus sp. differed among themselves (intergeneric differences) 6-8 enzymes.

We find the one different enzyme among Poecilia wingei.
It can speak about difference gentics between both species.
We are going to make DNA-analysis of both species for confirm it.
If it will confirm - it willl be different species.
